Fans
Damn, I would say it's a short. as fans normally run for around 30 seconds, but no more. I would say check the wiring, sometimes the fan wiring rubs and shorts out. If not plug in Toyota Techstream and have a look.

Not shifting into 5th gear
Don't put it in to 4, just leave it in D. As for the Shaft speed sensor looks like it needs changing. I take it from the year it's a RX/RS 300 Engine. Which if you tell them its a lexus RX/RS300 engine they should know. I Believe it's easy to change that sensor as it's on the rear side of the gearbox, but never actually changed one.

2.4 Alphard , heater stopped working
Here's how I bleed the system, Put the front end of the vehicle facing uphill, run the engine so the temp gets up, (with rad cap off). then keep squeezing the top rad pip and run the internal heating on full blast (hot as you can) on intake not circular, and eventually the air lock will go.

Headlights - main beam / auto etc
I never use auto lights, I found them to be buggy and the sensor often gets glogged up with dirt.

The only place to get a new one is from Toyota Direct which can ship from Japan, but be prepared to pay hundreds of pounds for one. Other than that you can try Aliexpress

Spare Keys for 30 Series Alphard ?
You do not need to get one from Japan, any key blade can be cut and the remote as long as it's the right immobiliser chip will work. Normally you can buy them from Ali express and have a local key place cut it and programme it.

Issue with rear sliding doors, gap and wind noise/draft
You mean the side doors or the boot? Tue boot cam be adjusted by undoing the bolts on the closing mechanism. The side doors should have guiders to secure them into and a auto pull close motor (this is not the same as the closing motor which can be operated by the driver, ots the final soft close motor) if both doors are the same then look and see you have a fuse gone. Normally it goes and the soft close stops causing a gap at the bottom of the door
